Naturforsch. 52c, 761-765 (1997); received July 30/August 19, 1997 Polyphenoloxidase, o-Diphenols, 4-Pentadecylpyridine, Competitive Inhibitor, Taphrina pruni 4-Pentadecylpyridine was isolated from plums infected by Taphrina pruni. This fungal me tabolite has been tested for its effect on polyphenoloxidase, using the polyphenoloxidasecatalysed addition of L-proline to 4-methylcatechol in a chromogenic assay The resulting data prove that 4-pentadecylpyridine is a competitive inhibitor to polyphenoloxidase with an inhibitor constant of 3.70 ± 0.03*10-4 m . Further comparative measurements have demon strated that the effect is due to the pyridine moiety. Probably reduced polyphenoloxidaseactivity decreases the toxicity of o-diphenols to the fungus and thus increases susceptibility of plums to the fungus Taphrina pruni.

Some Biochemical Properties of Polyphenoloxidase from Spearmint (Mentha arvensis)

Polyphenoloxidase (PPO; EC 1.14.18.1) extracted from Mentha arvensis leaves was isolated by (NH4)2SO4 precipitation and extensive dialysis. Its optimum pH and temperature varied with the substrate. The PPO showed activity with various diphenols. Km values were found 0.825, 0.928 and 7.41mM for caffeic acid, 4-methylcatechol and catechol, respectively. On heat-inactivation, half of the activity ...

Pyrophosphatase, Peroxidase and Polyphenoloxidase Activities during Leaf Development and Senescence.

Inorganic pyrophosphatase, peroxidase, and polyphenoloxidase activities were studied as the function of leaf insertion level in eight monocotyledonous and eight dicotyledonous species. Alkaline inorganic pyrophosphatase shows a declining activity toward the end of senescence whereas no regular drift in either peroxidase or polyphenoloxidase activities was noticed during senescence of attached l...

Study on Barley and Malt Polyphenoloxidase. I. Chromatography of Barley, Steeped Barley, and Malt Polyphenoloxidase

A comparative chromatography of polyphenoloxidase isolated from barley, steeped barley, and green malt was made. It consisted ofextraction, precipitation, ion-exchange chromatography, gel filtration, and electrofocusing. Polyphenoloxidase was resolved into a high-molecularweight enzyme and a low-molecular-weight one. The proportion of the two enzymes underwent variations during malting. An incr...

Development of a new amperometric biosensor based on polyphenoloxidase and polyethersulphone membrane*

An amperometric biosensor based on the enzyme polyphenoloxidase (PPO), which makes the bioelectrocatalysis of phenolic compounds, was developed and optimized using cathecol as substrate. Polyethersulphone membranes were used for enzyme immobilization. Polyphenoloxidase oxidizes monophenols (cresolase activity) and diphenols (catecholase activity) into the corresponding o-quinones; the o-quinone...

Competitive inhibition of copper amine oxidases by vitamin B hydrochloride in chickpea

Copper amine oxidases (CAOs) catalyse the oxidative de-amination of biogenic amines which are ubiquitous compounds essential for cell growth and proliferation. The enzymes are homodimers containing both topaquinone and a Cu(II) ions as cofactors at the active site of each subunit. After extraction and purification of chickpea (cicer arietinum) amine oxidase by chromatoghraphy, Km and Vmax of th...
